Introduction
Trypsin is a porcine pancreas-derived enzyme that is commonly used to cleave proteins holding the cultured cells to the dish, so that the cells can be removed from the plates. The working concentration of trypsin is generally 0.25%, while sensitive cells are often used with low concentration (0.05%) of trypsin. EDTA, a chelating agent, enhance the enzymatic activity by removing calcium and magnesium ions. These ions obscure the peptide bonds on which trypsin acts as well as enhancing cell to cell adhesion.
