Introduction
It is commonly used for the detection of protein on PVDF membrane, nitrocellulose membrane and cellulose acetate membrane; immerse the PVDF membrane, nitrocellulose membrane or cellulose acetate membrane with protein in the Ponceau staining solution and shake for 5-10 Minutes or more, after the protein bands appear on the membrane, take out the blotting membrane and rinse with distilled water, PBS or other appropriate solution for 2-3 times, each time for 3-5 minutes, until the red background disappears, the subsequent follow-up WB detection.
1. Staining of NC membranes: 0.2-2 % Ponceau S in 3 % trichloroacetic acid + 3 % sulphosalicylic acid, incubate for 1-5 mins. at room temperature. Enhance contrast by incubation in water acidified by a few drops acetic acid.
2. Staining of PVDF membranes: 0.1-1 % Ponceau S in 10 % acetic acid, incubate 1-5 mins. at room temperature. Ponceau S stained bands on PVDF only appear bright and distinct after drying.
